基于电催化氧化还原型谷胱甘肽的测定

摘要:研究了还原型谷胱甘肽( GSH) 在乙炔黑修饰碳糊电极(AB-CPE) 上以对乙酰氨基酚(PA)为媒介的电催化氧化行为。实验结果表明,GSH在CPE和AB-CPE电极上电化学活性较弱,对乙酰氨基酚(PA)对改善CPE和AB-CPE电极对GSH的电化学响应具有催化作用。采用计时电流法(CA) 、循环伏安法(CV) 、交流阻抗法(AC)和差分脉冲伏安法(DPV)研究了GSH 在AB-CPE 上的电化学;用DPV测得的电催化氧化峰电流值与溶液中的GSH浓度在7.5×10-7~1.0×10-4 mol/L范围内呈良好的线性关系(R=0.998),检出限(S/N=3,σ=4.81%)为9.1×10-7 mol/L。将该方法用于市售眼药水中还原型谷胱甘肽含量的测定,结果满意。

Determination of reduced glutathione based on electrocatalytic oxidation
Abstract:Reduced glutathione (GSH) was studied in acetylene black on the modified carbon paste electrode (AB-CPE) electric catalytic oxidation behavior. It was found that glutathione itself showed a poor electroche-mical response at CPE and AB-CPE, the electrochemical behaviors could be greatly enhanced by using PA, which enables a sensitive electrochemical determination of the substrate glutathione. Methods: chronoampero-metry(CA), Cyclic voltammetry(CV), AC impedance method(AC), Differential pulse voltammetry (DPV). The kinetic parameters of the reaction were evaluated. The catalytic oxidationpeak currents of glutathione versus its concentration had a good linear relationship in the concentration range of 7.5×10-7~1. 0×10-4 mol/L with the correlation coefficient of 0. 998,and the detection limit of 9.1×10-7 mol/L by DPV.Conclusion: AB-CPE can catalyze the oxidation of GSH well. Furthermore,The method used to detecter the content of reduced glutathione in commercial eye drops has received satisfying results.
