纳米壳聚糖骨形成蛋白水凝胶预防大鼠牙槽嵴萎缩的实验研究

摘要:目的:以纳米壳聚糖等为载体,加入骨形态发生蛋白,制备成水凝胶,注入大鼠拔牙窝观察其促进牙槽骨修复,预防牙槽嵴萎缩的效果。方法:选用SD大鼠54只,随机均分为3组,拔除大鼠下颌切牙,制备大鼠剩余牙槽嵴吸收模型。向实验组、对照组拔牙窝内分别注入纳米壳聚糖骨形成蛋白水凝胶、纳米壳聚糖水凝胶,空白对照组不植入任何物质。术后3、6、9周每组分别处死6只大鼠,完整分离下颌骨,高频钼靶X线摄影,图像用IPP软件分析测量。标本行大体观察和组织学观察。结果:在3、6、9周处死大鼠时,实验组的剩余牙槽嵴相对高度明显高于对照组和空白对照组,差异有统计学意义(p<0.05);病理切片显示实验组牙槽窝成骨情况优于单纯壳聚糖温敏性胶体组和空白对照组。结论:纳米壳聚糖骨形成蛋白水凝胶能有效预防拔牙术后剩余牙槽嵴萎缩。

A Study concerning the Effect of Nano-Chitosan/Bone Morphogenetic Protein Gel on Preventing Absorption of Rat's Alveolar Ridge
Abstract:Objecte:To investigate the effect of the Nano-Chitosan(NCS) bone morphogenetic protein (BMP) hydrogel injected into Extraction sockets of the rat on preventing absorption of residual alveolar ridge. Methods: The compound of the Nano-chitosan loading with recombinant bone morphogenetic protein 2,(rhBMP-2) where prepared. 54 SD rats were divided randomly and equally into 3 groups: the experimental group, the control group, and the blank group. Nano-Chitosan bone morphogenetic protein hydrogel (NCS/BMP Gel) were implanted into the rat's alveolar fossa of the experimental group ,Alveolar fossa of control group implanted with Nano-Chitosan hydrogel (NCS Gel), fossa of blank group left empty. Rats were separately sacrificed after 3,6,9 weeks.Molybdenum target X-ray examination, pathologic observation were evaluated. Results:The relative length of residual alveolar ridge in experimental group were predominant than those of other groups at 3,6,9 weeks after operation. A significant difference between experimental and control groups(p<0.05) were observed. Pathologic image showed that new bone formation in residual alveole ridge of the rate at 3,6,9 weeks after implantation, The condition of the new bone regeneration in the experimental group were better than those in the control group and blank group . Conclusion: NCS/BMP hydrogel can be used to prevent the absorption of residual alveolar ridge.
