低氧环境对内皮细胞BDNF表达的影响

摘要:目的:心脏微血管内皮细胞在心肌血管新生充当重要作用,低氧环境(5% O2)对内皮细胞脑源性神经营养因子(BDNF)表达的影响,目前所知甚少。方法:使用实时荧光定量PCR对经分离的大鼠心脏微血管内皮细胞,人EC304内皮细胞株在低氧环境下的不同时间点,BDNF 表达水平进行研究。结果:研究结果提示:随着低氧处理时间的延长,细胞死亡数明显增加。大鼠心脏微血管内皮细胞,低氧处理4h,BDNF表达没有明显变化,低氧处理8h后,BDNF表达明显增加,12h达到顶峰,24h后呈明显下降,且低于低氧处理8h后水平。而人EC304细胞株,低氧处理3小时后,BDNF表达呈现下降,于6h开始接近正常水平,12h后BDNF表达量显著升高,在24h后表达呈下降并低于正常水平。结论:低氧环境能促使心脏微血管内皮细胞死亡,同时能使内皮细胞的BDNF表达,在一定时间范围内呈现升高,于12小时达到高峰,但在24小时内其表达水平呈下降至接近正常水平。

BDNF Expression in Endothelial Cell during Hypoxia
Abstract:Objective: Cardiac microvascular endothelial cells (CMECs) play an important role in myocardial angiogenesis. However, the impact of low oxygen (5% O2) on brain-derived neurotrophic factor (BDNF) expression of endothelial cells is still not clear. Methods: Real-time PCR was applied to investigate the BDNF expression in CMECs and human endothelial cell line EC304 under hypoxic environment in different time points. Results: It was found that: 1) with the extension of hypoxic time, the number of dead cells increased significantly; 2) for CMECs, exposed to hypoxia within 4h, BDNF expression did not change significantly, however, BDNF mRNA expression was increased significantly in hypoxia 8h, and then arrived at the peak level in hypoxia 12h, decreased at hypoxia 24h; 3) for EC304, BDNF expression was decreased in hypoxia 3h, and arrived at normal level in hypoxia 6h, and then arrived at the peak level in hypoxia 12h, decreased at hypoxia 24h. Conclusion: Hypoxia (5% O2) was able to incur cell death of CMECs. In addition, hypoxia was able to increase expression of BDNF in CMECs. It was arrived at peak level in hypoxia 12h. However, it was decreased at hypoxia 24h.
