斑马鱼牙齿生长发育的观察研究

摘要:目的 观察斑马鱼牙齿的生长发育情况,探讨斑马鱼作为模式生物研究人类牙齿发育相关基因的可行性。方法 使用扫描电镜、HE切片染色以及茜素红染色等方法观察斑马鱼的牙齿生长。 结果 茜素红染色观察斑马鱼牙齿在4dpf时开始附着于腮弓,至第七天开始换牙,之后斑马鱼牙齿处于交替换牙状态;HE染色观察斑马鱼牙齿具有牙髓腔、牙本质等结构;扫描电镜观察成年斑马鱼牙齿整体状况,可观察到其牙齿没有牙根。结论 斑马鱼牙齿可作为人类牙齿发育模型进行相关基因功能研究,其中茜素红染色为观察早期4-6天斑马鱼牙齿发育的最佳方法,可结合显微注射技术研究牙齿发育相关基因的功能。

Study on the growth and development of zebrafish's tooth
Abstract:Objective To research the development of zebrafish's tooth, and to explore the feasibility of studying the gene function in human tooth development applying zebrafish's tooth development. Methods The tooth of zebrafish was observed by alizarin-staining, HE staining and the scanning electron microscope. Results Alizarin staining results showed that the attachment of zebrafish's tooth starts at 4dpf, and the first replacement tooth attaches at 7dpf, followed by complicated tooth replacement. The pulp cavity and dentine could be observed by the light microscope after HE staining of paraffin sections. Under the scanning electron microscope, the overall dentition of adult zebrafish can be observed. The teeth are presented no dental root. Conclusion Zebrafish's tooth could be used as a model organism of human tooth development research. The best method to observe early tooth development of zebrafish aged 4-6dpf is alizarin-staining, which can be used with the technology of microinjection to study the function of tooth development related genes.
