家蚕乙酰胆碱酯酶与谷胱甘肽-S-转移酶抗性与敏感性三级结构分析

摘要:本文对农药具有抗性及敏感性家蚕体内的乙酰胆碱酯酶(AChE)和谷胱甘肽-S-转移酶(GSTs)基因进行了基因扩增与克隆,根据AChE和GSTs基因翻译的氨基酸序列用同源模建方法和从头模建方法分析了抗性蚕品种与敏感性蚕品种蛋白氨基酸的突变对三级结构的影响。结果显示,抗性与敏感性蚕品种的AChE一共有4个氨基酸差异,分别是抗性18位的异亮氨酸、486的天冬氨酸、553的赖氨酸、606的缬氨酸突变为敏感的蛋氨酸、甘氨酸、谷氨酸和丙氨酸。抗性与敏感性蚕品种的GSTs只有1个氨基酸差异,即抗性93位的缬氨酸突变为敏感的丙氨酸。氨基酸的差异对其三级结构产生一定影响,从而影响酶活性,使家蚕的抗药性发生改变。

Analysis on the tertiary structure of acetylcholinesterase and glutathione-S-transferase in pesticide resistant and sensitive silkworm
Abstract:In this paper, the acetylcholinesterase (AChE) and glutathione-S-transferase (GSTs) genes of pesticide resistant and sensitive silkworm were amplified and cloned. The amino acid sequences and the tertiary structures of AChE and GSTs in accordance with the gene translation homology modeling were established according to the modeling methods. The results showed that a total of four amino acid differences of AChE between pesticide resistant and sensitive silkworm were screened, which 8 isoleucine, 486 aspartic acid, 553 lysine and 606 valine in resistant variety weere muted to methionine, glycine, glutamic acid and alanine in sensitive variety. For GSTs, there was only one amino acid difference between resistant and sensitive silkworm varieties, i.e., the 93 valine in resistance was changed into alanine in susceptibility. Amino acid differences produce an impact on its tertiary structure, thus affecting enzyme activity, so silkworm resistance was changed.
