NaCl刺激保加利亚乳杆菌对丙酮酸代谢及相关酶活性的影响

摘要:本文研究了NaCl刺激保加利亚乳杆菌对丙酮酸代谢及关键酶活性的影响,分析其与提高保加利亚乳杆菌冻干存活率的关系。用高效液相色谱法对葡萄糖、有机酸、乙酰辅酶A(Acetyl-CoA)以及胞内能量代谢变化进行了分析,顶空气相色谱法对乙醛进行测定,分光光度法和比色法对关键酶活性进行了分析。NaCl刺激保加利亚乳杆菌对丙酮酸的代谢影响较大,葡萄糖代谢速率提高48.18%(P<0.01),乳酸生成速率提高8.93%(P<0.01),关键中间代谢产物Acetyl-CoA的含量约增加31.35%(P<0.01);而乙酸含量出现负增长,乙醛生成速率降低了22.86%(P<0.05)。胞内ATP代谢变化明显促进糖代谢速率;乳酸脱氢酶、乙酸激酶、磷酸乙酰转移酶的活性均有增加,而乙醛脱氢酶活性减弱。结果表明,NaCl刺激保加利亚乳杆菌对自身糖代谢的加快,丙酮酸代谢碳流向的重新分配以及关键酶活性的改变,可能与提高保加利亚乳杆菌的冻干存活率相关。

Effect of NaCl stress on pyruvate metabolism and enzyme activity in Lactobacillus bulgaricus
Abstract:The aim of this study was to investigate the effects of NaCl stress on metabolism of pyruvate and activity of key enzymes in Lactobacillus bulgaricus, and to analyze the relationship between NaCl stress and survival rate of freeze-dried. Glucose, organic acids, acetyl coenzyme A (Acetyl-CoA) and intracellular energy metabolism were analyzed by high performance liquid chromatography (HPLC); Acetaldehyde was determined by headspace gas chromatography (HS-GC); activity of Key enzymes were mearsured by spectrophotometry and colorimetry. Pyruvate metabolism of Lactobacillus bulgaricus was significantly changed by NaCl stress; The utilization rate of glucose and the production rate of lactate were increased by 48.18% and 8.93% respectively (P<0.01), the content of key intermediate metabolite Acetyl-CoA increased 31.35% approximately (P<0.01);the content of acetic acid demonstrated negative growth, and the production rate of acetaldehyde was decreased 22.86% (P<0.05).Changes of intracellular ATP metabolism was contributed to accelerate the utilization of glucose; the activities of lactate dehydrogenase, acetate kinase, and phosphate acetyltransferase were increased by NaCl stress, the activity of aldehyde dehydrogenase was decreased.The results show that,The survival rate of freeze-dried was increased may be related to the increase of glucose utilization, rerouting of pyruvate metabolism, and the changes of the key enzyme activities in Lactobacillus bulgaricu by NaCl stress.
