意大利苍耳叶总酚的提取及抗氧化活性研究

摘要:【目的】以意大利苍耳叶为原料,研究其总酚的提取条件,同时对意大利苍耳叶总酚提取物的抗氧化活性进行评价。【方法】选用乙醇作为溶剂, 采用超声辅助提取法处理意大利苍耳叶。应用中心组合设计和响应面分析相结合的方法,模拟得到回归方程的预测模型和可信度,并通过岭脊分析得到最佳提取条件。使用福林酚试剂对意大利苍耳叶的总酚含量进行测定。分别采用铁离子还原法、DPPH自由基清除法测定意大利苍耳叶提取物的抗氧化能力。【结果】超声波辅助提取意大利苍耳叶总酚的最佳工艺条件为:乙醇浓度54%、超声时间31 min、溶媒比195 mL/g,意大利苍耳叶总酚提取率为3.12±0.27%。意大利苍耳叶总酚提取液对铁离子还原的IC50为774.5±7.26mg药材/L, 对DPPH自由基的半数抑制率IC50为250.9±5.48mg药材/L。【结论】由响应面法优化得到的意大利苍耳叶总酚的提取工艺方便可行,得到的意大利苍耳叶总酚具有一定抗氧化活性。

Total phenol extraction process optimization and antioxidant activity of the leaves of Xanthium italicum Moretti
Abstract:【Objective】In this study, the extraction of phenolic compounds from leaves of X. italicum Moretti and the total phenol extract antioxidant activity were investigated. 【Method】 Using ethanol-water mixture as solvent, the leaves of X. italicum Moretti were processed by ultrasonic assisted extraction method. A central composite design and response surface methodology were used to optimize experimental conditions for extraction of phenol. The total phenolic content was determined by the Folin-phenol reagent. Antioxidant capacity was measured using iron reduction method and DPPH radical scavenging method respectively. 【Results】The optimum conditions of ultrasonic assisted extraction were ethanol concentration 54%, extraction time 31 min, liquid-solid ratio 195 mL/g, and the yield of total phenolic was 3.12±0.27%. The leaves of X. italicum Moretti on the reduction of iron ions IC50 was 774.5±7.26 mg medicinal material weight /L and DPPH IC50 was 512.4±6.48 mg medicinal material weight /L.【Conclusion】X. italicum Moretti leaves which are obtained by the response surface method to optimize the extraction process of total phenol convenient and feasible, the X. italicum Moretti leaves total phenol has certain antioxidant activity.
