摘要:目的:探讨强制性运动疗法(constraint-induced movement therapy, CIMT) 是否能够促进脑缺血的神经发生并抑制凋亡。方法:成年大鼠随机分为三组:正常组、缺血组、缺血后给予CIMT组。使用内皮素制作脑缺血模型。脑缺血后1周使用石膏绷带固定健侧上肢,进行为期3周的CIMT。使用免疫荧光染色检测DCX和NeuN的表达,使用TUNEL法检测细胞凋亡。结果:侧脑室下区的新生神经细胞在脑缺血后明显增多。CIMT能够进一步促进侧脑室下区新生神经的增殖及海马齿状回新生细胞的增殖并抑制了凋亡。结论:CIMT能够促进成年大鼠脑缺血后的神经发生。

Effects of constraint-induced movement therapy on neurogenesis after stroke
Abstract:Objective: To investigate whether constraint-induced movement therapy (CIMT) could influence ischemia-induced neurogenesis, apoptosis in the adult rats. Methods: Adult rats were divided into a sham group, an ischemia group, and an ischemia treated with CIMT group. Ischemia was induced by injection of endothelin-1(ET-1). Rats of CIMT group were treated with a plaster cast to restrain the healthy forelimb for 3 weeks beginning 1 week after ischemia. The expression of DCX and NeuN were measured by immunohistochemistry, and apoptosis was measured by TUNEL assay. Results: The production of neuroblasts in the subventricular zone (SVZ) was significantly increased after stroke. CIMT enhanced the proliferation of newborn neurons in the SVZ as well as newborn cells in the dentate gyrus. Furthermore, CIMT suppressed apoptosis in the adult rats. Conclusion: CIMT is effective in enhancing neurogenesis in the adult rats after stroke.
