金属离子对黄酮类化合物与牛血清白蛋白之间反应的协同作用研究

摘要:本文采用荧光光谱法研究了三种黄酮类化合物山萘酚-3-O-α-L-鼠李糖基-(1→3)-?-L-鼠李糖基-(1→6)-β-D-吡喃半乳糖苷(A1)、山萘酚-7-O-鼠李糖基-3-O-芸香糖苷(A2)和山奈甲黄素-7-O-4"-O-乙酰基-鼠李糖基)-3-O-芸香糖苷(A3)与牛血清白蛋白之间的相互作用时,共存离子存在的条件下,离子对反应的协同作用,研究表明,在295K的条件下,Ca2+和Fe3+对药物A1和A3存在负协同作用,在310K条件下,Fe3+对药物A1和A3存在负协同作用,除此之外,其余的金属离子均对三种药物与蛋白质之间的作用存在正协同作用,并通过对反应的热力学常数的计算,对作用方式进行了衡量, 在T=295,310 K情况下Ca2+ -BSA-A1,Al3+-BSA-A2,Al3+-BSA-A3,Fe3+-BSA-A3体系,是典型的疏水作用力,在T=295 K 时,Al3+-BSA-A1,Cu2+-BSA-A3,主要为静电作用力,在其他的情况下,均是ΔS<0时,主要是氢键和van der Waals力起作用。

Hill equation was used to study the quenching mechanism of bovine serum albumin by flavonoids with presence of common ions.
Abstract:IThe cooperative effects of common ions on the interactions between three kinds of flavonoids and BSA were studied by Hill equations. The flavonoids were obtained from the leaves of Actinidia kolomikta(Rupr.et Maxim.)Planch. As the results shown, most of the common ions have positive cooperative effects on drugs-BSA system. However, at 295K, Ca2+ and Fe3+ have negative cooperative effects on drug A1 and A3-BSA system, at 310K, Fe3+ has negative cooperative on drug A1and A3-BSA system. The interaction forces between flavonoids and BSA were also invested in the presence of ions. At 295, 310 K, In Ca2+ -BSA-A1,Al3+-BSA-A2,Al3+-BSA-A3 and Fe3+-BSA-A3 systems, ΔH>0 and ΔS>0, hydrophobic interaction plays a key role; At 295 K, in Al3+-BSA-A1 and Cu2+-BSA-A3 systems,ΔH<0,ΔS>0, electrostatic interaction plays a key role; Other conditions, the ΔS<0, hydrogen binding and van der Waals force play a major role.
