响应曲面法优化鹿角盘胶原蛋白ACE抑制肽的酶解工艺研究

摘要:以鹿角盘胶原蛋白为原料,用胰蛋白酶酶解制备ACE抑制肽。采用高效液相色谱(HPLC)测定鹿角盘胶原蛋白酶解液的ACE抑制率,同时以ACE抑制率和水解度(DH)为指标,通过单因素试验及响应曲面试验设计优化胰蛋白酶酶解鹿角盘胶原蛋白的酶解工艺。结果表明:各因素对ACE抑制率影响的主次顺序依次为酶解温度>pH值>酶解时间>加酶量,对DH抑制率影响的主次顺序依次为酶解温度>pH值>酶解时间>加酶量。胰蛋白酶酶解制备ACE抑制肽最佳酶解工艺为pH 8、加酶量4 %、酶解温度55 ℃、酶解时间4 h。在此基础上,鹿角盘胶原蛋白酶解液ACE抑制率82.07 %、DH 22.78 %。

Optimization of Enzymatic Hydrolysis of the Angiotensin-Ⅰ-Converting Enzyme(ACE) Inhibitory Peptides from Antler Base of Collagen by Response Surface Methodology
Abstract:The enzymatic preparation of angiotensin I-converting enzyme (ACE) inhibitory peptides from antler base of collagen using trypsin was optimized by response surface methodology, it was shown that degree of ACE inhibitory activity and hydrolysis(DH) of antler base of collagen hydrolysate, ACE inhibitory activity was measured by HPLC. The results showed that the factors to the ACE inhibitory activity was descended as hydrolysis temperature, pH value and hydrolysis time, enzyme loading, the DH was descended as hydrolysis temperature, pH value and hydrolysis time, enzyme loading. Ph8, enzyme loading 4 %, hydrolysis temperature 55℃and hydrolysis time 4 h were found to be the optimal conditions to obtain ACE inhibitory activity and DH can be reached to 82.07 %、22.78 % respectively.
