利福平作为神经氨酸酶抑制剂的研究

摘要:目的:流感是危害人类健康的重大疾病,目前临床一线抗流感药物都是以病毒释出环节为靶位的神经氨酸酶抑制剂。本论文通过高通量筛选评价成药库,寻找具有抑制流感病毒神经氨酸酶活性的化合物。方法:构建多株系流感病毒神经氨酸酶酶学水平模型;应用流感病毒神经氨酸酶模型对成药库进行筛选和评价。结果:我们成功构建了多株系流感病毒神经氨酸酶酶学模型,通过对成药库的高通量筛选,发现利福平有显著抑制神经氨酸酶活性的作用。结论:利福平有抗神经氨酸酶活性的作用,可为新型抗流感药物研究提供思路。

Study of Rifampin as neuraminidase inhibitor
Abstract:Objective: Influenza causes significant human illness and death. Neuraminidase inhibitors which target at viral release are the most frequently used clinical anti-flu drugs. Here, through screening of a FDA-approved drug library, drugs were assessed to search for new neuraminidase inhibitors. Methods: Multiple influenza neuraminidase enzymatic models were constructed and used to screen the drug library. Results: Through screening of the drug library, Rifampin was found to have inhibitory effect of influenza viral neuraminidase. Conclusion: Rifampin could effectively inhibit the activity of multiple influenza viral neuraminidase and could accumulate information for the study of new anti-flu drugs.
