摘要:基于精氨酸的胍基与邻二酮的特异性反应,设计合成了对维生素c代谢产物具有高选择性和灵敏度的近红外荧光探针(Arg-Cy)。该探针能与维生素c代谢产物L-木酮糖(L-xylosone)特异性反应,成功用于常氧条件下药理剂量的维生素c诱导人肝癌HepG2细胞死亡过程中细胞内L-xylosone含量变化的实时监测,并发现乏氧条件下药剂量的维生素c不能代谢产生L-xylosone。

A Near-infrared Fluorescence Probe for Monitoring the Metabolic Products of Vitamin c in living Cells
Abstract:Based on the reaction of the guanidine group in arginine (Arg) with adjacent diketone, we developed a near-infrared fluorescent probe (Arg-Cy) for monitoring the metabolic products of vitamin c. As a result, the probe can respond to L-xylosone, a metabolite of vitamin c, with high selectivity and sensitivity, and was successfully used to visualize the real-time changes of L-xylosone level in living cells. Furthermore, the probe can also be used to distinguish the different metabolic states of vitamin c in the process of pharmacological doses of vitamin c inducing HepG2 cell death under normoxic conditions and hypoxic conditions. The result showed that vitamin c can be metabolized to L-xylosone only under normoxic conditions due to the oxidative stress, but not under hypoxic conditions.
