芹菜素诱导乳头状甲状腺癌细胞自噬的研究

摘要:芹菜素是一种天然的植食性黄酮,它广泛地存在于多种水果和蔬菜中,还被公认具有抗炎症、抗氧化和抗癌的作用。通过实验研究了芹菜素对乳头状甲状腺癌(PTC)BCPAP细胞系自噬水平的影响。结果表明,芹菜素的浓度为50 μM时浓度依赖地强烈抑制BCPAP细胞的增殖,但却诱导提高了自噬的水平。实验通过检测一些自噬的标志性关键分子表达水平来表征自噬,包括LC3、p62和Beclin1等,另外还用AO染色通过流式细胞术来研究酸性自噬泡的形成。综上所述,本研究证实了在抑制甲状腺乳头状癌BCPAP细胞增殖的现象之下,芹菜素激发了BCPAP细胞中自噬的发生。

Study on apigenin induced autophagy in thyroid cancer cells
Abstract:Apigenin, a natural edible plant flavone which abundantly occurs in many fruits and vegetables, is recognized as a flavonoid with anti-inflammatory, antioxidant and anticancer properties. In this study, we investigated the effects of apigenin on the level of autopahgy in papillary thyroid carcinomas (PTC) cell line BCPAP. The results show that apigenin inhibited, in a dose dependent manner, BCPAP cell proliferation, with strong inhibition occurring at 50μM; and induced autophagy level as shown by the expression of several molecular markers such as LC3, p62, and Beclin 1. Furthermore, AO staining were performed to probe the formation of autophagic vacuole in apigenin-treated BCPAP cells. In summary, our results demonstrate that apigenin inhibits viability of BCPAP cells and simultaneously stimulates autophagy.
