不同剂量白藜芦醇对高脂饲喂小鼠非酒精性脂肪肝的影响

摘要:探讨不同剂量白藜芦醇对高脂膳食诱导的小鼠非酒精性脂肪肝的影响。6周龄C57BL/6雄性小鼠,随机分成4组:正常日粮组(ND)、高脂日粮组(HF)、高脂日粮+0.04%白藜芦醇组(HFL)和高脂日粮+0.08%白藜芦醇组(HFH)。饲喂17周处死小鼠,显微镜观察肝脏组织切片,测定血脂和肝脏脂类水平及脂代谢相关基因表达。与正常组相比,HF组血浆TG、TC、LDL-C和ALT、AST水平显著升高;与0.08%白藜芦醇相比,0.04%白藜芦醇可以显著降低小鼠血浆TG、TC、LDL-C和ALT含量。RT-PCR结果显示HF组小鼠Fas、SREBP1c、ACC的表达显著上调,AMPK、SIRT1、β2-AR、CPT1的表达显著下调;与HF组小鼠相比,HFL小鼠Fas、SREBP1c、ACC的表达显著降低,AMPK、SIRT1、β2-AR、CPT1的表达显著升高;0.08%白藜芦醇对于AMPK、SIRT1、Fas、SREBP1c、β2-AR和CPT1表达的影响与0.04%白藜芦醇相比具有显著性差异。因此,实验结果表明对于高脂膳食引起的脂代谢紊乱和非酒精性脂肪肝,0.04%白藜芦醇的改善效果好于0.08%的剂量,这可能是由于白藜芦醇发挥调节脂类代谢的作用与剂量有着密切的关系。

Effects of different doses of resveratrol on nonalcoholic fatty liver disease in mice fed with high-fat diet
Abstract:To investigate the impacts of different doses of resveratrol (RSV) on lipid metabolism in high-fat diet (HFD) induced nonalcoholic fatty liver disease (NAFLD) in mice. Six-week-old C57BL/6 male mice were fed with normal diet (ND), HFD control (HF), HFD+0.04% RSV (HFL, 0.04% RSV with HFD, w/w) and HFD+0.08% RSV (HFH, 0.08% RSV with HFD, w/w). Mice were sacrificed after 17 weeks. Liver histological analysis was performed by microscopy. Plasma and hepatic lipid profile and lipid metabolism related genes were measured. Compared with the ND group, the levels of TG, TC, LDL-C and ALT, AST were significantly elevated in the plasma of HF mice. 0.04% RSV was more effective than 0.08% RSV in reducing the levels of TG, TC and ALT. RT-PCR results showed that the expressions of Fas, SREBP1c and ACC were significant up-regulated, while AMPK, SIRT1, CPT1 and β2-AR were significant down-regulated in HF mice. Compared with the HF mice, HFL mice had significant lower expressions of Fas, SREBP1c and ACC, and significant higher expressions of AMPK, SIRT1, β2-AR and CPT1. 0.08%RSV had remarkable impact on the expressions of AMPK, SIRT1, Fas, SREBP1c, β2-AR and CPT1 compared with 0.04%RSV. Thus, these results indicated that 0.04% RSV was more effective than 0.08% RSV on lipid metabolic disorders caused by HFD. This may be due to the effects of RSV on lipid metabolism were closely related to the dosage of RSV.
