体外培养骨骼肌卫星细胞适宜电刺激模型的建立

摘要:目的:体外培养骨骼肌卫星细胞(C2C12),建立模拟运动的适宜电刺激模型。方法:培养小鼠C2C12细胞,利用C-Pace EP电刺激仪按不同电压、频率、刺激时间、脉冲时间施加电脉冲,MTT法检测C2C12细胞活性。结果:与对照组相比,电压≤10 V、刺激时间≤150 min、频率≤5.0 Hz、2 ms≤脉冲时间≤4 ms时,各实验组吸光度无统计学差异。结论:电刺激C2C12细胞时,其条件以电压≤10 V、刺激时间≤150 min、频率≤5.0 Hz、2 ms≤脉冲时间≤4 ms为宜,细胞活性不受影响。

The establish on skeletal muscle satellite cells model in vitro of the suitable electrical stimulation
Abstract:Objective: To establish a model of the suitable electrical stimulation through culturing skeletal muscle satellite cells (C2C12). Method: C2C12 cell was cultured and stimulated by C-Pace EP according to different Voltage, Time, Frequency and Duration. Cell activity of C2C12 cell was determined by MTT method, and the protein content of MYH in C2C12 cell was detected by Western blot. Result: Comparing with the control, when Voltage≤10 V, Time≤150 min, Frequency≤5.0 Hz, 2 ms≤Duration≤4 ms, there was no statistical difference. Conclusion: when exert electric stimulation on C2C12 cells, its suitable condition: Voltage≤10 V, Time≤150 min, Frequency≤5.0 Hz, 2 ms≤Duration≤4 ms.
