转录因子YY1在氡致细胞凋亡中的作用

摘要:目的 探究转录因子YY1在氡致细胞凋亡中的作用,为研究氡致细胞凋亡的分子机制提供基础资料。方法 以BEAS-2B细胞为研究对象,将细胞暴露于20000Bq/m3氡浓度的气体染毒装置中,染毒30min。连续染毒2次作为染毒1代(Rn1)。分别染氡5代(Rn5)、10代(Rn10)、15代(Rn15)、20代(Rn20)。流式细胞术检测细胞凋亡率,Western blot检测YY1的表达水平。构建YY1过表达载体和沉默载体,分别转入BEAS-2B细胞中观察PARP-1、Bcl-2和Bax表达水平的改变。结果 YY1的表达水平随氡染毒剂量的增加而降低,而细胞凋亡率随氡染毒剂量的增加而增加。细胞转染YY1过表达载体后,PARP-1和Bcl-2表达上调,Bax表达下调,细胞凋亡率降低。结论 氡致BEAS-2B细胞凋亡率的增加,可能是通过YY1调控凋亡相关基因的表达实现的。

YY1 modulates radon induced apoptosis in BEAS-2B cells
Abstract:Objective To explore the role of YY1 in radon induced apoptosis and to provide experimental data for mechanistic study. Methods BEAS-2B cells directly exposed to radon(20000Bq/m3) twice for 30 mins were defined as 1st generation (Rn1), and continued for Rn5, Rn10, Rn15, and Rn20. The expression levels of YY1 were determined by Western blot, and cell apoptosis was determined by flow cytometry. The PcDNA3.1-YY1 and shRNA-YY1 were transfected into the cells to observe changes in expressions of PARP-1, Bcl-2 and Bax. Result Upon radon exposure, the apoptosis rate in BEAS-2B cells increased with a down-regulation of YY1 expression. In the cells transfected with over-expressed YY1 plasmid, the apoptosis rate decreased with a down expression of Bax and up expression of Bcl-2 and PARP-1. Conclusions The increased apoptosis in radon induced cells might be mediated by YY1 and its target genes of PARP-1 and Bax/Bcl-2.
