临床乳腺炎奶牛MAdCAM-1、VCAM-1蛋白表达差异分析

摘要:乳腺炎是奶牛业最具危害性的疾病之一,其发病率高、损失大,若不及早治疗会导致奶牛过早淘汰,因此给奶牛产业发展造成了严重影响。黏膜地址素细胞黏附分子MAdCAM-1与α4β7 结合可以介导淋巴细胞归巢,血管细胞黏附分子VCAM-1与α4β7 结合,调节淋巴细胞参与免疫应答。本研究以MAdCAM-1和VCAM-1为切入点,应用Western Blot技术,分析MAdCAM-1、VCAM-1 在健康奶牛与临床乳腺炎奶牛乳腺组织中蛋白的表达差异;应用流式细胞技术,探究临床乳腺炎奶牛及健康奶牛乳汁中整合素家族α4β7的阳性率,从而进一步探明奶牛乳腺黏膜归巢地址素 MAdCAM-1、VCAM-1 的蛋白表达规律,为乳腺炎的治疗提供研究依据。

Analysis on protein expression differences of MAdCAM-1 and VCAM-1 on clinical mastitis of dairy cattle
Abstract:Mastitis as one of the most harmful diseases in dairy industry seriously effects the development of dairy industry, which will cause dairy being eliminated at early stage for its high incidence as well as huge losses. MAdCAM-1 known as mucosal addressin cell adhesion molecules, can be combined withα4β7 to mediate lymphocyte homing. Vascular cell adhesion molecule abbreviated to VCAM-1 can be combined withα4β7 to adjust lymphoid cells in the immune response. The present study took MAdCAM-1 and VCAM-1 as the starting point to analysis the differences of MAdCAM-1 and VCAM-1 expression between clinical mastitis mammary gland and healthy breast by using Western Blot analysis. To explore the positive ratio ofα4β7 between clinical mastitis mammary gland and healthy breast by using flow cytometry (FCM) analysis, which helps to explore the expression of MAdCAM-1 as well as VCAM-1 and has laid an experiment foundation for further study on mammary gland immunity.
