白藜芦醇促进Nrf2磷酸化保护UVA诱导的HaCaT细胞损伤

摘要:目的:探讨白藜芦醇(resveratrol,Res)保护UVA辐射造成正常人皮肤角质细胞HaCaT氧化损伤的分子机制,为临床预防和治疗UVA辐射及皮肤癌提供新的理论依据。方法:UVA和Res分组处理HaCaT细胞,相差显微镜下观察细胞的形态改变;MTT法检测细胞活性;流式细胞术分析细胞内ROS含量, Western blot分析核内、外Nrf2蛋白含量。结果:1. 治疗组和预防组中Res均能明显改善细胞形态和活性;2. UVA辐射后,HaCaT细胞DCF平均荧光强度由对照组14.7增至26.4,Res治疗组和预防组分别下降为16.4和18.8;3. 白藜芦醇能够促进Nrf2进入HaCaT细胞核。4. Nrf2入核可能跟其自身磷酸化有关。结论:Res能够通过促进Nrf2入核激活Nrf2,进而保护UVA辐射诱导HaCaT细胞内氧化应激损伤。

Resveratrol on the protection of HaCaT cells from UVA-induced damage through promoting the phosphorylation of Nrf2
Abstract:Objective: The aim of this study was to investigate the protective role of resveratrol in human keratinocytes (HaCaT) against UVA-induced oxidative damage and the possible mechanism, so as to provide new theory for prevention and therapy UVA radiation and skin cancer in clinic. Methods: The HaCaT cells were UVA irradiated and the effects of resveratrol on cell morphous, cell viability and reactive oxygen species (ROS) generation were measured used inverted microscope, MTT and flow cytometry, respectively. The proteins of Nrf2 inside and outside HaCaT cells were determined by western blot. Results: 1. Cell viability was improved obviously in both therapy and prevention groups. 2. After UVA irradiation, mean fluorescence intensity increased from 14.7 (control group) to 26.4 (UVA group) in HaCaT cell, but the number was 16.4 and 18.8 in therapy and prevention groups, respectively; 3. resveratrol facilitated Nrf2 accumulation in the nucleus; 4. the accumulation in the nucleus of Nrf2 may be related with phosphorylation of itself.Conclusion: Our results suggest that resveratrol facilitate Nrf2 accumulation in the nucleus, thereby protecting HaCaT cells from UVA-induced oxidative stress.
