基于CO1基因3′端序列对15种蚜小蜂的分子鉴定

摘要:蚜小蜂科寄生蜂是粉虱、介壳虫、蚜虫等农林害虫重要天敌,在害虫生物防治中发挥重要作用。目前,蚜小蜂分类鉴定仅依靠传统分类的方法远远无法满足需要,迫切需要一种方法能够快速准确识别蚜小蜂。本研究利用在线粒体CO1基因3′端分子标记,采用DNA条形码分析方法研究蚜小蜂的分类鉴定技术,旨在探究该分子标记鉴定蚜小蜂的可行性,以期建立一种快速准确鉴定蚜小蜂的新方法。结果显示:扩增的CO1基因3′端序列长626bp,A+T的含量都在75%左右,表现出明显的T+A偏向性;碱基转换与颠换的比值为0.36。从系统发育树的聚类分析来看,每种蚜小蜂都能形成单独分支。从种间和种内遗传距离来看,COI基因3'端序列种间遗传距离是种内的30.8倍,该分子标记都能很好的对蚜小蜂进行分类鉴定。?????

Study on Molecular Identification of 15 aphelinids based on partial sequences of the CO1 gene
Abstract:The species of Aphelinidae are the important natural enemies of whiteflies, aphids and scale insects. Many of them play an important role in biological control of pests. Currently, it is a more difficult task for taxonomy identification only by morphological characteristics. Developing a new method to identify species more effectively is urgently needed. This study aimed to make molecular identification analysis of common species of Aphelinidae by 3′-end of mt DNA CO1 gene. The analysis method is DNA barcoding technology. The results are as follows: The average length of CO1 sequences was 626bp. A + T content was about 75%, showing significant T+A bias; the ratio of abasic transition to transversion was 0.36. each aphelinid formed a separate branch according to the cluster analysis of the phylogenetic tree. The interspecific genetic distance was 30.8 times of the intraspecific genetic distance of sequence of 3′- end. 3′-end of mt DNA CO1 gene are effective molecular markers to identify species of Aphelinidae.?????
