黄莲内生真菌的分离及其抑菌活性的研究

摘要:背景:药用植物内生菌是筛选生物活性成分或先导化合物的有效途径之一,Cc-3是从药用植物黄莲中分离出的内生真菌,探讨Cc-3对植物病原菌的抑制作用,为植物病害的生物防治奠定基础。方法:采用对峙法和对扣法对Cc-3进行玉米大斑病菌、核盘菌、烟曲霉、尖孢镰刀菌、稻瘟菌、玉米灰斑病菌、蓖麻枯萎病菌、番茄灰霉病菌等植物病原真菌的抑菌活性测定;利用顶空固相微萃取(HS-SPME)气相色谱-质谱(GC-MS)联用法鉴定菌株Cc-3产生的挥发性活性物质;通过形态学和分子生物学对Cc-3菌株进行鉴定。结果表明Cc-3对多种病原真菌具有较强的抑菌活性,尤其对核盘菌和玉米大斑病菌抑制效果较好;菌株Cc-3产生的挥发性活性物质为四甲苯;经鉴定该菌株为多喙茎点霉菌(Phoma multirostrata)。结论:从药用植物黄莲中分离出多喙茎点霉菌Cc-3对核盘菌和玉米大斑病菌抑制效果较好,是潜在的植物病害的生物防治菌株。

Isolation of Endophytic Fungi from Coptis chinensis Franch and Screening Activities Against Pathogenic Fungi
Abstract:Background: The endophytes of medicinal plants are one of the effective ways for screening bioactive ingredients and leading compounds. The strain of Cc-3 is endophytic fungi isolated from the medicinal plant Coptis chinensis and we are exploring the inhibition effect of Cc-3 against the plant pathogens, laying a foundation for the biological control of plant disease. Methods: By using the methods of confrontation and clasping for the determination of antifungal activity about Cc-3 against Setosphaeria turcica, Sclerotinia sclerotiorum, Aspergillus fumigatus, Fusarium oxysporum, Magnaporthe Oryaze, Cercospora zeae-maydis, Fusarium oxysporum, Cylindroca-rpon destructans and Botrytis cinerea and so on. Using headspace solid phase microextraction (HS-SPME), gas chromatography and mass spectrometry(GC-MS) is used to identify the volatile active substances by Cc-3. The strain of Cc-3 was identified by morphological and molecular biological methods. The results show that the strain of Cc-3 has strong antifungal activity against a variety of plant pathogens, especially Setosphaeria turcica and Sclerotinia sclerotiorum and the volatile active substance is tetramethylbenzene producing; The strain of Cc-3 was Phoma multirostrata.Conclusion: The Cc-3 strain of Phoma multirostrata isolated from the medicinal plant Coptis chinensis had better inhibition effect against etosphaeria turcica and Sclerotinia sclerotiorum and it is a potential biological control strain of plant disease .
