棉纤维品质相关蔗糖合酶基因SuSA1功能标记开发

摘要:蔗糖合酶基因SuSA1是纤维发育的调节因子,与纤维品质紧密关联。本研究以蔗糖合酶基因SuSA1为候选基因,分别从纤维品质一般的陆地棉中棉所8号和优质纤维海岛棉Pima90-53中扩增该基因,得到SuSA1在A亚组的DNA序列SuSA1-1和在D亚组的DNA序列SuSA1-2。依据SuSA1-1在陆海种间SNP变异,设计了标记引物S1-1-11,依据SuSA1-2在海岛棉中存在12bp的碱基缺失,设计标记引物S1-2-1。利用本课题组前期构建的[(CCRI8×Pima90-53)×CCRI8] BC1群体的连锁遗传图谱,对开发的标记进行连锁分析,发现S1-1-11、S1-2-1分别位于连锁群A8、D8上,结合纤维品质数据,对开发的标记进行纤维品质性状QTL定位,当LOD值为5.0时,A8 S1-1-11处存在马克隆值的QTL,可解释表型变异的11.1%,当LOD值为3.0时,在D8 S1-2-1处存在马克隆值QTL,可解释表型变异的9.7%。这将为棉花分子标记辅助育种及纤维品质改良提供重要标记资源。

Development of Functional Markers of Sucrose Synthase Gene SuSA1 Related to Fiber Quality in Cotton
Abstract:Sucrose synthase gene SuSA1 SuSA1 is an important regulatory factor of fiber development of and closely linked with fiber quality in cotton. In this study, based on the SuSA1, A-genome sequence of SuSA1 named SuSA1-1 was obtained from Gossypium hirsutum CCRI8 with medium fiber quality and G. barbadense Pima90-53 with good fiber quality, and D-genome sequence of SuSA1 named SuSA1-2 was obtained from the two cottons. The markers S1-1-11 and S1-2-1 were designed according to the SNP difference of SuSA1-1 between CCRI and Pima90-53 and deletion of 12 base pairs in the Pima90-53 of SuSA1-2. Based on the genetic linkage map in the BC1 population of (CCRI8×Pima90-53)×CCRI8 constructed by our lab, two markers S1-1-11 and S1-2-1 were mapped at A8(c8) and D8(c24), respectively. Combined with the previous fiber quality data, QTL mapping for fiber quality traits were performed. QTL related to microniare was mapped at A8 with LOD value of 5.0 and associated with the marker S1-1-11. This QTL could explain 11.1% of phenotypic variation. With LOD value of 3.0, the linkage group D8 contained QTL related to microniare, and this QTL explained 9.7% of phenotypic variations. This will provide the marker resource for molecular marker-assisted selection and improvement of fiber quality in cotton.?????
