摘要:目的 水通道蛋白-1(AQP1)是表达于腹膜并参与调节腹膜功能的重要分子,其表达水平和生物学功能对于腹膜透析非常重要。研究pH值对AQP1启动子区域活性的影响。方法:通过Real-time PCR检测pH值对HEK-293T细胞AQP1 mRNA表达水平的影响。通过构建含有AQP1潜在启动子区域的荧光素酶报告基因载体pGL3-AQP1 Promoter-4K,检测pH值对该区域活性的影响。结果:HEK-293T细胞AQP1 mRNA水平的表达随pH值上升而增加。AQP1上游0至-4000bp的区域活性受pH值的调控。结论:AQP1上游0至-4000bp的区域对于pH值调控AQP1表达非常关键。

The effect of pH value on activity of AQP1 promoter
Abstract:Objective To study the effect of pH value on activity of AQP1 promoter region. Method Using real-time PCR, the effect of pH value on expression of AQP1 mRNA was measured. A luciferase reporter construct containing fragment from -4000 to +1 bp region upstream of AQP1 was cloned into the pGL3-basic vector, and luciferase activity was measured after transfection of this construct into HEK 293T cells. Result The AQP1 mRNA expression was upregulated by the increase of pH value in HEK-293T cells. The active regulatory element affected by pH value, was observed in the -4000bp~0bp region upstream of AQP1. Conclusion The -4kp region upstream of AQP1 was of chief importance on pH value regulating AQP1 expression.
