太行菊组织培养前种子消毒方法

摘要:消毒剂对植物种子消毒效果及萌发都有一定影响,为了探索太行菊种子的最佳消毒方法,本试验选用0.1%氯化汞和10%H2O2做为消毒剂,分别经过两种消毒剂处理后接种到1/2MS培养基上,通过比较其污染率和发芽率得出最佳消毒方法:先将种子在无菌水中浸泡4个小时左右,然后用75%酒精迅速漂洗15s,之后用1%的氯化汞浸泡8min(加两滴吐温-20),最后用无菌水冲洗2次~3次后将种子接至1/2MS培养基上。结果表明,消毒最佳的是0.1%氯化汞处理10min,污染率为0;发芽率最佳的是0.1%HgCl2消毒6min;0.1%氯化汞消毒效果要比10%H2O2消毒效果好。

The seed disinfection method of Opisthopappus taihangensis tissue culture
Abstract:Disinfectant has some influences on disinfection and germination of plant seed. To find the best disinfection method of Opisthopappus taihangensis (Ling) Shih seed, 0.1%HgCl2 and 10%H2O2 are used as two kinds of disinfections in this paper. The seeds are inoculated in 1/2MS culture medium after treated by the two disinfections. By comparing pollution rate and germination percentage of the two different disinfection methods, the best disinfection method is found. Specifically, the steps of the best method are as follows: Firstly, the seeds are soaked in sterile water for 4h or so. Secondly, the seeds are treated quickly by 75% for 15s. Thirdly, 0.1%HgCl2(add two drops of twain-20) is used for 8min. Finally, Opisthopappus taihangensis (Ling) Shih seeds are inoculated to 1/2MS culture medium after washed with sterile water for 2~3 times. The result shows that, the method, which is 0.1%HgCl2 6min, can thoroughly disinfect the seeds with 0 pollution rate; and 0.1%HgCl2 6min treatment is the best germination percentage choice; the disinfection effect of 0.1%HgCl2 is better than 10%H2O2 .
