理化因素对运动发酵单胞菌絮凝的影响及絮凝机理的研究

摘要:本工作研究了20种理化因素对运动发酵单胞菌Zymomonas mobilis ZM401絮凝的影响。结果表明:ZM401的絮凝物质在酸性环境中易降解而在碱性环境中较稳定,并且对温度不敏感。金属阳离子的添加对ZM401的絮凝无显著影响,其中镁离子浓度的增加会使絮凝率微弱提高2.9%。单糖的添加对絮凝几乎没有影响,蔗糖和纤维二糖的增加会提升絮凝率。有机溶剂的添加会微弱增加ZM401的絮凝,其中乙醇有大约2.6%的增幅,说明絮凝物质间的结合受疏水作用的微弱影响。ZM401的絮凝率在α-淀粉酶、壳聚糖酶、脂肪酸酶、蛋白酶K添加时无显著变化,但其对纤维素酶非常敏感,0.15 FPU/mL的纤维素酶即可使絮凝颗粒不可逆的解体。上述实验结果证明了运动发酵单胞菌的絮凝物质是纤维素类物质,其相互作用为简单的分子间物理搭桥,对环境变化不敏感,能为细胞提供稳定的絮凝状态。

The effect of physical and chemical factors on the flocculation of Zymomonas mobilis and the flocculation mechanism research
Abstract:The effect of 20 kinds of physical and chemical factors on the flocculation of Zymomonas mobilis ZM401 has been studied in this work. The results showed that: the flocculation material of ZM401 could be easily degraded in acid, but was stable in alkaline environment. Moreover, the flocculation was not sensitive to temperature. The addition of metal cations had no obvious effect on the flocculation, only with the increasing of Mg2+, the flocculation efficiency had a slightly increasing by 2.9%. Monosaccharide had little effect on the flocculation, while sucrose and cellobiose had postive effect on the flocculation efficiency. The addition of organic solvents increased the flocculation, and ethanol improved the flocculation efficiency by about 2.6%. The addition of the α-amylase, chitosanase, lipase and protease K almost had no influence on the flocculation while flocculation was very sensitive to cellulase, which with 0.15 FPU/mL caused the flocculation irreversible collapse. It is proved that the material related to flocculation of Zymomonas mobilis is cellulose, by which cell adhered to each other.
