芋螺毒素V超家族序列特征分析

摘要:芋螺毒素V超家族是一类目前已知数量较为稀少的超家族,紫霞芋螺中V超家族数目在鉴定到的所有芋螺毒素超家族总数目中所占比例的绝对优势让我们进一步的对这个超家族进行关注。通过对该家族翻译序列、5'和3'端非翻译区序列以及半胱氨酸密码子的同源性的分析,V超家族在目前已知物种间的同源性非常高。通过与另一个同样拥有XV与VI/VII号半胱氨酸框架的O2超家族序列的比较,两者的XV号框架应该是趋同进化导致,并非来源于同一祖先。让人惊讶的是,我们在V超家族的3'UTR区域首先发现重复序列的存在,这一(TATC)n重复序列在不同的物种间也是高度保守,且目前为V超家族的独有特征。

Sequence Analysis of V-conotoxins Reveals Repeat Domain at 3' Untranlated Region
Abstract:V-conotoxin is so far a rare superfamily, however, 16 out of 69 sequences identified in conus flavidus belongs to this superfamily, which highly attached our interest on this superfamily. By homologous analysis of the translated region, 5' and 3' untranslated region (UTR), and cysteine codon in the cysteine framework, we found V-conotoxins share high sequence homologue in different species. Although both V and O2 superfamilies bear both XV and VI/VII cysteine frameworks, the cysteine codon applied in the same position of the framework is different, which may indicate the convergent evolution of the same cysteine framework in these two superfamilies. To our interest, it was the first time that identification of repeat domain at the 3' UTR of V-contoxins. The (TATC)n repeat in the 3' UTR is conserved in different species, and so far is a unique feature for V superfamily.
