ALDH1A3表达与脑胶质瘤的间质表型及预后的关系

摘要:目的:探索乙醛脱氢酶 1A3(aldehyde dehydrogenase 1A3, ALDH1A3)在间质型(Mesenchymal, Mes)脑胶质瘤中的表达,以及其表达与脑胶质瘤的恶性程度和生存期的关系。方法:利用大规模临床样本,通过mRNA 芯片、mRNA测序和相关统计学方法验证ALDH1A3 基因表达量与间质型脑胶质瘤的关系及与患者生存期的关系。结果:ALDH1A3的表达与脑胶质瘤的恶性程度相关,在高级别脑胶质瘤中ALDH1A3高表达患者预后较差。在胶质母细胞瘤分子病理分型中,ALDH1A3在间质型脑胶质瘤中表达最高,可以作为间质型脑胶质瘤的特异性标志物。结论:ALDH1A3表达水平可用于判断脑胶质瘤患者的预后,还可以作为Mes型脑胶质瘤较理想的分子标志物,并且其表达与肿瘤的恶性程度和生存期显著相关。

Aldehyde dehydrogenase 1A3 forebodes mesenchymal glioma and predicts poor prognosis
Abstract:Aim: The aim of this study was to investigate the expression of ALDH1A3 in mesenchymal glioma. And its effect on the WHO grade and prognosis of glioma. Methods: By whole-genome transcriptome microarray and mRNA sequencing analysis, we compared the expression of ALDH1A3 in high- and low- grade gliomas as well as different molecular subtypes. Relevant statistical methods validated the relationship between ALDH1A3 expression and prognosis of mesenchymal glioma. Results: High ALDH1A3 mRNA expression was related to mesenchymal phenotype and poor clinical outcomes in gliomas. ALDH1A3 can serve as a reliable marker of Mes subtype gliomas. Conclusion: Our study demonstrate that ALDH1A3 overexpression is significantly associated with high grade as well as the higher mortality of high-grade gliomas in survival analysis. ALDH1A3 could also be a novel marker for Mes subtype gliomas.
