基于鱼胶原蛋白的组织工程角膜基质载体支架的构建及鉴定

摘要:目的:以鳗鲡鱼皮胶原为材料,通过交联的方法构建组织工程角膜载体支架,评价其理化性能、组织结构及生物相容性。方法:本文利用鳗鲡鱼皮胶原蛋白为材料,通过1-乙基-3-(3-二甲基氨基丙基)碳二亚胺(EDC)/N-羟基丁二酰亚胺(NHS)交联冻干的鱼皮胶原蛋白制备组织工程角膜载体支架,鉴定其外观、透光率等理化性质,HE染色和扫描电镜检测其组织结构。接种本实验室建立的非转染无致瘤性人角膜基质(HCS)细胞系细胞,检测胶原载体支架的生物相容性。结果:实验结果表明,制备的组织工程角膜具有良好的光学性能和均一的组织结构。胶原支架浸提液对HCS细胞没有毒性作用,且利于细胞生长增殖。接种的HCS细胞粘附于胶原载体支架中,在支架中分布较为均匀,表明构建的鱼皮胶原蛋白载体支架具有良好的生物相容性。结论:以上结果表明制备的组织工程角膜支架具有良好的理化性质和生物学功能,有望用于组织工程角膜的构建。

Construction of Tissue-engineered Corneal Stroma Scaffold by Fish Skin Collagen and its Characterization
Abstract:Purpose: Collagen from skin of Anguilla japonica is used to construct tissue-engineered corneal scaffold by cross-linking. Physical and chemical property and biocompatibility of the collagen scaffold were measured and analyzed. Methods: The fish collagen purified from the skin of Anguilla japonica was used to construct collagen scaffold with N-(3-Dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride crystalline (EDC)/N-Hydroxysuccinimide (NHS) as cross-linking agents after collagen freezing-drying. Physical and chemical property of collagen scaffolds were identified, including transparency and histological structure by HE staining of frozen sections and scanning electron microscope. Besides, human corneal stroma (HCS) cells were cultured for the research of collagen scaffold biocompatibility. Results: Collagen scaffold presented good transparency, uniform reticular organizational structure. The extracted solution from collagen scaffolds was not toxic to HCS cells, but was suitable for their growth and reproduction. After inoculation, the HCS cells immigrated into the scaffold and distributed regularly, indicating collagen scaffold had good biocompatibility to HCS cells. Conclusion: The tissue-engineered corneal scaffold had good physical and chemical property and biocompatibility, could promisingly be applied in vitro in the construction of tissue-engineered cornea.
