摘要:目的:探讨沉默信息调节因子3对肝癌细胞增殖的影响。方法:western blot检测SIRT3在正常肝组织、永生化肝细胞及肝癌细胞系中的蛋白表达水平;在肝癌细胞中过表达SIRT3,台盼蓝排斥实验检测过表达SIRT3对肝癌细胞增殖的影响,Brdu标记实验检测SIRT3对肝癌细胞DNA合成的影响;在肝癌细胞中过表达SIRT3,平板集落实验检测SIRT3对肝癌细胞集落形成能力的影响;裸鼠异位移植瘤模型中验证SIRT3对肿瘤生长的影响。结果:SIRT3在肝癌细胞系中的蛋白水平较永生化肝细胞、正常肝组织降低;过表达SIRT3抑制肝癌细胞的增殖,抑制肝癌细胞DNA合成;过表达SIRT3抑制肝癌细胞的集落形成能力;过表达SIRT3在裸鼠异位移植瘤中抑制肝癌细胞的生长。结论:SIRT3可抑制肝癌细胞的增殖。

Effect of SIRT3 on proliferation of hepatocellular carcinoma
Abstract:Objective: To illuminate the effect of SIRT3 on the proliferation of hepatocellular carcinoma. Methods: The protein expression of SIRT3 in a panel of two normal liver tissue, two immortalized hepatocyte, and three hepatoma cell lines was determined by western blot. The overexpression of SIRT3 was conducted by transfection of vector expressing SIRT3.Proliferation of HCCs was assayed by trypan blue exclusion assay. DNA synthesis was tested by BrdU incorporation assay. The ability of colony formation was analysed by colony formation assays. The effect of SIRT3 on proliferation of HCC was investigated by xenograft tumor model in nude mice. Results: SIRT3 expression was significantly lower in three hepatoma cell lines than the others. SIRT3 overexpression lead a marked decrease in cell number as tested by trypan blue exclusion assay. BrdU incorporation assay revealed that SIRT3 overexpression reduced DNA synthesis. SIRT3 overexpression reduced the number and size of cell colonies as determined by colony formation assays. Nude mice were used to investigate that SIRT3 inhibited the proliferation of HCC. Conclusion: SIRT3 inhibit the proliferation of HCCs.
