摘要:高效的遗传转化体系是研究植物基因功能的基础。目的:建立根癌农杆菌注射法介导的丹参遗传转化体系。方法:以丹参叶片为外植体,潮霉素作为筛选压,利用农杆菌注射法对丹参叶片进行了遗传转化研究。结果:通过GUS组织化学染色和PCR鉴定分析,所得再生植株均为转基因阳性株系。结论:农杆菌注射丹参叶片可显著提高转基因株系的获得率,为丹参基因功能研究及突变体库的建立奠定了基础。

Establishment of genetic transformation system of Salvia miltiorrhiza by agroinfiltration
Abstract:Efficient transformation system is the basis for gene function studies. This paper presents the establishement of agroinfiltration-mediated transformation of Salvia miltiorrhiza. We used the leaves as explants for Agrobacterium infiltration. The results showed that all the plantlets regenerated on selection medium containing high hygromycin concenstration are positive as examined by genomic PCR and GUS staining. It will provide basis for Salvia miltiorrhiza gene function studies and mutant library construction.
