茶树β-葡萄糖苷酶基因及其对UV-B诱导的响应

摘要:β-葡萄糖苷酶(bGlu)可以水解茶叶中结合态的香气成分,促进茶叶挥发性化合物释放,提高茶叶香气。本研究以茶树叶片UVB-SSH文库获得的差异cDNA片段为基础,利用RACE技术从'福鼎大白茶'叶片中克隆了bGlu基因 cDNA全长,共2194bp,包含1887bp的开放阅读框,可编码628个氨基酸,具有β-葡萄糖苷酶特征功能区,属于糖苷水解酶第三家族。该基因编码的β-葡萄糖苷酶定位于质膜,属亲水蛋白,具跨膜区。qRT-PCR分析表明,适宜剂量的UV-B辐射能够诱导茶树叶片bGlu基因表达上调,但过量的UV-B辐射会损伤叶片细胞,进而抑制基因表达。

β-Glucosidase gene in tea plant and its expression induced by UV-B irradiation
Abstract:β-Glucosidase (bGlu) can hydrolyze the volatiles bound with glycosides to release aromatic compounds, resulting in enhance of tea aroma. A full length cDNA of bGlu gene was cloned by RACE and RT-PCR from tea leaf (Camellia sinensis) based on a differentially expressed cDNA in the UVB-induced tea leaf subtractive hybridization cDNA library. The cDNA was 2194bp in length, with an ORF 1887 bp encoding a protein with 628 amino acids. The protein encoded by the bGlu gene had a functional domain of β-glucosidase, belonging to glycoside hydrolase III family. It locates in the plasma membrane, belonging to hydrophilic protein with a transmembrane region. The qRT-PCR test revealed that expression of the bGlu was stimulated by an appropriate dose of UV-B irradiation, but was suppressed by overdose of UV-B.
