摘要:基于微液滴的体外区室化-荧光激活细胞分选(in vitro compartmentalization-fluorescence-activated droplet sorting, IVC-FACS)技术是对酶进行筛选和进化的有力手段。IVC-FACS技术能够在皮升级微液滴中实现单细胞酶活性超高通量检测分选,通过荧光底物来反映酶活性的高低,因而要求产物荧光分子在液滴必须有良好的保留性才能满足筛选要求。本研究以亲水性良好的7-羟基香豆素-3-羧酸(7-Hydroxycoumarin-3-carboxylic acid,HCCA)作为离去基团,合成了新型的磷酸三酯酶荧光底物。相比于文献报道的基于的3-氰基-7-羟基-4-甲基香豆素 (3-Cyano-7-hydroxy-4-methylcoumarin,CHMC)荧光底物,其催化产物的液滴保留性大大提高。进一步用新型底物对GkaP/26A8进行超高通量模式分选表明,活性细胞能从大量的背景细胞中有效富集,最高富集效率超过900倍。证实了该底物在磷酸三酯酶超高通量筛选中的应用潜力。

New Generation of Fluorogenic Substrate for Ultrahigh Throughput Screening of Phosphotriesterases
Abstract: In vitro compartmentalization based fluorescence-activated droplet sorting (IVC-FACS) is a power tool for enzyme high throughput screening. It is crucial for an IVC-FACS system to keep the stability of the fluorescent signal inside a particular microreactor, so that the information for the reaction progress can be readily maintained during several hours of screening. Coumarin derived fluorogenic substrates are commonly used for the detection of many enzymes. However, the rapid diffusion of coumarin between droplets severely hampered its application in IVC-FACS. Here, a hydrophilic carboxylic acid group modified coumarin, 7-Hydroxycoumarin-3-carboxylic acid (HCCA), was reported with good retention in droplet. An HCCA-phosphotriester substrate was synthesized for the IVC-FACS screening of phosphotriesterase. The new substrate could diffuse into the w/o/w double emulsion droplets and react with the enzyme, producing fluorescent product which could be confined well in droplets. Also, the enzyme activity could be faithfully reflected in droplet assays. The model screening assay revealed that enzyme-expressing E.coli cells could be effectively enriched as high as 900-folds from large accesses of background cells, further demonstrating the suitability of the HCCA derived substrate in the application of IVC-FACS system.
