MiRNA-BD:基于证据的microRNA生物标志物识别模型

摘要:MicroRNA(miRNA)与生物体内许多重要的生物过程相关。大量研究表明,miRNA可以作为疾病临床诊断和治疗的有效生物标志物。目前,miRNA生物标志物的识别主要依赖于生物学实验筛选以及生物信息学算法预测。这两类方法中,前者需要较大的时间和成本开销,后者模型往往缺乏通用性。直到现在,还没有任何普适性的规则以用于miRNA生物标志物的发掘。基于对miRNA-mRNA调控网络子结构的分析,发现了miRNA生物标志物识别的两个重要证据并且构建了相应的通用识别模型。与传统基于miRNA协同调控机制的方法相比,该模型更加关注miRNA独立调控基因以及调控具有重要生物功能基因的能力。目前,该模型已经在MiRNA-BD软件中实现并且成功应用至一些复杂疾病miRNA生物标志物的识别中。

MiRNA-BD: an evidence-based model for microRNA biomarker discovery
Abstract:MicroRNAs (miRNAs) are associated with many important biology processes and plenty of studies have been reported on detecting miRNA as biomarkers for clinical diagnosis and treatment. Most of the approaches are experimental, which are timing consuming and costly. Bioinformatics models are therefore preferred and developed. However, until now, very few models are evidence-based and no general rules can be applied to biomarker discovery. Based on sub-structural analysis of miRNA-mRNA interaction network, two evidences are uncovered and considered into a novel evidence-based model for miRNA biomarker discovery. Compared to routine methods which focus on miRNA synergic functions, the model pays more attention to miRNAs' independent regulatory power and their biological functions. The model is implemented in the program MiRNA-BD and its application in biomarker discovery for several diseases demonstrated its predictive power.
