摘要:本文基于荧光共振能量转移原理,设计合成了一种新型罗丹明-丹磺酰衍生物(DRh)。通过使用UV-Vis和荧光光谱,考察了探针DRh对于金属离子的识别作用。结果表明,探针DRh对Hg2+具有良好的选择性。结合Job's点和电喷雾质谱(ESI-MS),证明了DRh和Hg2+形成了化学计量比1:1的配合物。在0-30 μmol L-1范围内,荧光强度比(I582 nm / I538 nm)与Hg2+的浓度成正比,Hg2+检出限为0.008 μmol L-1。而且,该探针被成功的应用于活体HeLa细胞中Hg2+的成像研究。

A new fluorescent probe for Hg2+ based on FRET and its application to living cells imaging
Abstract:On the basis of fluorescence resonance energy transfer, a new dansyl-rhodamine derivative (DRh) was synthesized. The recognition properties of the probe DRh with metal ions had been investigated by the UV-Vis and fluorescence spectrophotometry. The results showed that the probe DRh exhibited the selective recognition of Hg2+. The 1:1 stoichiometric structure between DRh and Hg 2+ were supported by Job's plot and ESI-MS. The linearly fluorescence intensity ratio (I582/I538) is proportional to the concentration of Hg2+ in the range 0-30 μmol L-1. The limit of detection of Hg2+ is 0.008 μmol L-1. The present probe was applied to the fluorescence imaging for living HeLa cells.
