华癸中慢生根瘤菌7653R ctrA基因组成型表达的共生表型及宿主互作蛋白的初步筛选

摘要:本文首先以pBBR1MCS-5为载体构建重组质粒,将额外拷贝ctrA基因导入华癸中慢生根瘤菌7653R。表型检测结果显示,其不影响根瘤菌7653R在培养条件下的生长速度,但在共生条件下导致宿主植物紫云英结瘤数量下降、根瘤固氮活性降低。通过荧光定量PCR技术,发现ctrA基因在根瘤菌培养与共生根瘤中均呈组成型表达,并且外源质粒的导入使得ctrA在根瘤中的表达量稍有增加。另外,以CtrA为诱饵,筛选紫云英根瘤AD-cDNA酵母双杂交文库,获得4个阳性克隆。序列分析表明其中一个候选的靶蛋白为JUN 激酶激活域结合蛋白。

Symbiotic phenotypes of constitutive-expressed ctrA in Mesorhizobium huakuii 7653R and screening for its host interacting protein
Abstract:By using the vector pBBR1MCS5, an extra copy of ctrA was introduced into Mesorhizobium huakuii 7653R. The phenotypes were examined for both free living cells and symbiosis. It was found that the growth rate of 7653R free-living cells remained unchanged, but nodule numbers in host plant Astragalus sinicus were decreased and nodule nitrogen fixation ability was reduced as well. Real time qPCR analyses showed that ctrA was constitutively expressed under cultivation and symbiosis conditions, its expression level was slightly increased due to the introduction of pBBR-ctrA plasmid. Meanwhile, basing a yeast two-hybrid system screening in Astragalus sinicus AD-cDNA library, four candidate clones which interacted with bait protein CtrA were obtained. Sequencing analyses indicated that one of the cDNA inserts encoded a JUN kinase activation domain binding protein.
