三个长穗偃麦草矮秆种质系的鉴定和分析

摘要:长穗偃麦草(Thinopyrum elongatum, syn. Lophopyrum elongatum, Agropyron elongatum)是小麦重要的近缘植物之一。本研究利用细胞学和原位杂交技术对三个小麦-长穗偃麦草后代选育的矮秆种质系进行了鉴定,结果表明矮杆种质系山农31504 染色体数目为44,细胞学稳定,属于二体异附加系;山农31504-1和山农31504-3 染色体数目为42,细胞学稳定;以拟鹅冠草(St)总基因组DNA为探针进行基因组原位杂交鉴定,结果表明山农31504附加了2条长穗偃麦草St基组染色体,山农31504-1和山农31504-3为小麦-长穗偃麦草小片段易位系;利用pTa535和pSc119.2为探针对山农31504-1、31504-3和鲁麦5号的双色荧光原位杂交,发现山农31504-1和31504-3的6B、1D染色体发生相同的结构变异,说明二者是相同的小麦-长穗偃麦草渐渗系,染色体结构变异可能发生在6B、1D染色体上。?????

Identification and analysis of a novel dwarfing gene in three Thinopyrum elongatum germplasm lines
Abstract:Thinopyrum elongatum is an important relatives of wheat. We intended to indentify three dwarfing germplasm lines which were selected from filial generation of wheat and Thinopyrum elongatum by cytology and in situ hybridization. As the results indicating that the number of chromosome of Shannong31504 is 44 and it has cytological stability, which indicate that it is a disomic alien addition line; the number of chromosome of Shannong 31504-1 and 31504-3 are 42 and they also have cytological stability. GISH was proceed using genomic DNA from Pseudoregnaria strigosa(St) as the probe concluding that the two alien chromosomes of Shannong31504 belonged to St genome, Shannong31504-1 and 31504-3 were wheat-Thinopyrum elongatum alien translocation with little fragmentation of Thinopyrum elongatum. FISH with the probe of pTa535 and pSc119.2 was used to establish the genome constitution of Shannong 31504-1 and 31504-3. We found that their 6B and 1D chromosomes have the same structural mutation. The results indicateing that they are the same wheat-Thinopyrum elongatum introgression lines, and the structural mutation may occur on 6B and 1D chromosomes.?????
