二穗短柄草低分子量谷蛋白亚基基因的分子特征

摘要:二穗短柄草(Brachypodium distachyon L.)作为一个新的禾谷类植物的模式种,针对其籽粒蛋白质组分的研究还不够深入。本研究借助蛋白质组学和分子遗传学手段对短柄草籽粒中谷蛋白进行了详细研究。通过和普通小麦的比较分析表明,两者的整体蛋白质图谱比较相似,但是短柄草籽粒中醇溶蛋白和高分子量谷蛋白亚基(HMW-GS)缺乏,清蛋白、球蛋白和低分子量谷蛋白亚基(LMW-GS)的丰度比较高,特别是C型LMW-GS比普通小麦要高。核酸杂交(Southern-blotting)表明短柄草中含有4-5个LMW-GS基因拷贝。另外利用位点特异PCR(AS-PCR),我们从短柄草中克隆得到18个LMW-GS基因。蛋白质杂交(Western-blotting)和质谱(MALDI-TOF-MS)鉴定技术进一步验证了这18个LMW-GS以及4个预测氨基酸序列。系统进化分析表明短柄草和山羊草(Ae. markgrafii和Ae. umbellulata)的亲缘关系要比小麦更近一些。

Molecular characterization of low molecular weight glutenin subunit genes in Brachypodium distachyon L.
Abstract:Brachypodium distachyon L. is a newly emerging model plant system for temperate cereal crop species. However, its grain protein compositions are still not clear. In the current study, we carried out a detailed proteomics and molecular genetics study on grain glutenin proteins in B. distachyon. SDS-PAGE and RP-HPLC analysis showed that Brachypodium has few gliadins and high molecular weight glutenin subunits, but possessed similar wheat electrophoretic patterns in albumins, globulins and low molecular weight glutenin subunits (LMW-GS) compositions. In particular, the LMW-C type subunits in Brachypodium were more abundant than these in common wheat. Southern blotting analysis confirmed that Brachypodium has 4-5 copies of LMW-GS genes. A total of 18 LMW-GS genes were cloned from Brachypodium by allele specific PCR. LMW-GS and 4 deduced amino acid sequences were further confirmed by using Western-blotting, MALDI-TOF-MS, respectively. Phylogenetic analysis indicated that Brachypodium was closer to Ae. markgrafii and Ae. umbellulata than to T. aestivum.
