哺乳期大鼠甲状腺功能动态变化的研究

摘要:目的 观察大鼠不同哺乳期时期的甲状腺功能动态变化趋势。方法 选择成年Wistar大鼠100只,其中雌性78只,雄性22只,体重210-250g。将雌性大鼠随机分为6组:正常未孕组(N)、哺乳第5天组(L5d)、哺乳第10天组(L10d)、哺乳第15天组(L15d)、哺乳第20天组(L20d)和断乳第5天组(W5d),每组13只。所有动物常规饲料喂养,饮用自来水,实验期间所有动物自由进食饮水。除正常未孕组外将其他5组雌鼠与雄性大鼠按3:1合笼交配,分别在产后哺乳第5天、第10天、第15天、第20天和断乳第5天时股动脉取血,应用放射免疫法检测血清T3、T4、FT3、FT4、TSH。结果 FT3、FT4组间比较差异有统计学意义(P均 < 0.05)。T3、T4、TSH组间比较差异均无统计学意义(P均 > 0.05)。FT3和FT4在不同哺乳时期均有下降趋势,尤其在哺乳第5天时较正常未孕组下降最明显(P <0.05),在断乳第5天时与正常未孕组相近;TSH在不同哺乳时期均有升高趋势,同样在哺乳第5天时较正常未孕对照组升高最明显(P >0.05),在哺乳第20天和断乳第5天时与正常未孕组相近。在不同哺乳时期均有一定比例的大鼠FT3、FT4低于正常参考值范围,在哺乳5第天时此比例最高;在哺乳第5天时,有1例大鼠FT3、FT4值均低于正常参考值下限而TSH高于正常参考值上限。结论 在哺乳早期血清FT3、FT4较低,可能出现出现甲状腺功能失代偿现象,应加强哺乳期甲状腺功能监测,以便及早发现甲状腺疾病。

Dynamic changes of thyroid function in lactation rats
Abstract: Objective To observe the dynamic trend of thyroid function in various stages of lactation rats. Methods 100 Wistar rats (78 famale and 22 male),weighting 210-250 were selected. 78 adult female Wistar rats were randomly divided into 6 groups according to their weight: normal non-pregnant group(N), lactating the 5th day group(L5d), lactating the 10th day group(L10d), lactating the 15th group(L15d)and lactating the 20th day group(L20d)and weaning the 5th day group(W5d),13 rats in each group. All rats were fed with conventional fodder and the tap water freely, and the rats of lactating groups except the normal non-pregnant group cohabited with male rats (3:1). Then the rats in all groups were killed on the 5th day, 10th day, 15th day and 20th day after lactation and on the 5th day after weaning to get the blood respectively. The radioimmunoassay was used to characterize the T3、T4、FT3、FT4、TSH. Results FT3、FT4of all groups showed significant differences(all P < 0.05) ,T3、T4、TSH of all groups were not different, and the differences were not statistically significant(all P > 0.05). but FT3 and FT4 presented the downward trend in different lactation, decreasing obviously especially on the 5th day after lactation (P <0.05), and then recovered to normal level on the 5th day after weaning; TSH showed increasing trend in various stages of lactation rats, and similarly TSH increased most obviously on the 5th day after lactation compared with the normal non-pregnant group (P >0.05). Then TSH recovered to the normal level on the 20th day after lactation and the 5th day after weaning. FT3 and FT4 of a certain percentage of rats were below the normal reference range, in the different lactation period, with the highest proportion on the 5th day after lactation; on the 5th day after lactation, FT3, FT4 of one rat were lower than the lowest reference value, while TSH was higher than the highest reference value. Conclusions FT3 and FT4 were lower in early lactation, which may be a phenomenon of thyroid dysfunction. At the same time the monitoring of thyroid function should be strengthened for early detection of thyroid disease.
