大豆疫霉菌诱导的酵母双杂交文库构建及评价

摘要:大豆疫霉根腐病是危害大豆生产的毁灭性病害之一,研究利用SMART技术成功构建了疫霉菌诱导的大豆抗病品种绥农10的酵母双杂交文库。经检测得到的原始文库库容量高达1.2×106个单克隆,文库中插入的片段集中在0.5 ~ 1 kb,平均插入长度为700 bp。文库质量较好,能够通过筛选文库得到与目的蛋白互相作用的蛋白质。

Construction and Evaluation of Yeast Two-hybrid cDNA Library of Soybean Inoculated with Phytophthora sojae
Abstract:Phytophthora root and stem rot of soybean (Phytophthora sojae) is one of destructive diseases endangering soybean production, but there have been few reports on gene expression after inoculated with pathogen. Yeast- two- hybrid library of resistance soybean cultivar 'Suinong 10' inoculated with P. sojae race 1 was constructed successfully in this study. The results of detection showed that the library contained 1.2 ×106 independent clones. The sizes of most inserts were from 700 to 1000bp and average size of inserts of 600bp in the library. These results indicated that the library is suitable for screening on potential partner(s) of proteins of interest.
