摘要:为了增强环糊精固定相在高效液相色谱中的手性识别性能,本论文通过3-氯-4-甲基苯基异氰酸酯对正电型环糊精进行全取代,进而采用点击反应将环糊精共价键合至硅胶上制的一种新型的手性固定相。该固定相在甲醇的流动相中对黄烷酮类对映体具有很好的拆分能力,部分对映体的分离度超过了7.0。并且通过调节流动相的pH值引入了静电吸引作用,有效地拆分一些酸性药物。因此,设计的正电型环糊精的手性固定相有望实现广谱地拆分中性及酸性药物。

Clicked preparation of functionalised cationic cyclodextrin-based chiral stationary phases
Abstract:For the purpose of improving the chiral recognition ability of cyclodextrin-based stationary phases in high performance liquid chromatography (HPLC), this work firstly performed the persubstitution of cationic cyclodextrin with 3-chloro-4-methyl-phenylisocyanate. Subsquently, the functionalized cyclodextrins were anchored to silica via click reaction to afford a novel chiral stationary phase (CSP). This CSP exhibited excellent enantioseparations ability towards flavonoids with methanol-based mobile phase, the chiral resolutions of several enantiomers could exceed 7.0. Meanwhile, some acidic enantiomers could also be resolved by adjusting the Ph to construct the electrostatic interactions between cationic cyclodextrin and analytes. Thus, the well designed cationic cyclodextrin-based CSP is promising to realize the wide enantioseprations of neutral and acidic drugs.
