基于LC-MS/MS系统的羊绒羊毛检测方法研究

摘要:随着各种造假技术的发展羊绒羊毛的准确客观检测显得尤为重要,本文主要探究LC-MS/MS技术对羊绒羊毛纤维鉴别的可行性。基于蛋白质组学原理,通过还原法提取角蛋白并用胰蛋白酶对其进酶切,得到分子量较小的肽溶液,最后用Q Exactive 组合型四极杆 Orbitrap 质谱仪进行检测。结果表明,羊绒、羊毛、绵羊绒均具有各自特有的多肽片段,三者之间区别很明显,根据特征多肽片段可以准确客观的进行羊绒、羊毛、绵羊绒的鉴别。

Research of cashmere detection method based on the LC-MS/MS technology
Abstract:with the development of all kinds of fraud technology of cashmere, accurate and objective detection is particularly important. In this paper, we explore the the feasibility of cashmere wool fiber identification using LC - MS/MS technique . On the basis of the principle of proteomics we use reduction method to extract keratin and use trypsin to get smaller peptide solution and finally use Q Exactive combination quadrupole Orbitrap mass spectrometer for testing. The results showes that both cashmere, wool, sheep down have their unique peptide fragments, according to the characteristics of the peptide fragments cashmere, wool and sheep down can be accurate and objective identified.
