GABA通过抑制内质网未折叠蛋白反应减弱酒精对肝细胞的增殖抑制作用

摘要:抑制性神经递质Gamma-氨基丁酸(gamma-aminobutyric acid,GABA)在体内具有多种生理功能。有文献报道GABA可抑制酒精的肝毒性作用,但其机制不清。未折叠蛋白反应(Unfolded protein response,UPR)是细胞通过一系列信号转导途径维持内质网稳态的重要机制,但若UPR持续时间较长而使ER稳态不能及时重建,UPR信号通路将引发细胞凋亡。本研究采用MTT实验和Western blot等方法,探讨了GABA是否通过内质网非蛋白折叠反应发挥对抗酒精对肝细胞的增殖抑制作用。结果发现,酒精对体外培养的BRL肝细胞的增殖抑制作用可被GABA逆转,且酒精诱导的UPR标志物GRP78和CHOP表达上调被GABA抑制。以上结果说明GABA抑制酒精的肝毒性作用与GABA抑制酒精诱导的UPR有关。

Protective role of GABA against ethanol hepatotoxicity involves inhibition of unfolded protein response
Abstract:Gamma-aminobutyric acid (GABA), which is a primary inhibitory neurotransmitter, plays various physiological roles throughout the body. It was reported that GABA protects hepatocytes against ethanol-induced hepatotoxicity but the underlying mechanism is unclear. Using MTT and Western Blot analysis, the cytoprotective effect of GABA against ethanol-induced hepatotoxicity was evaluated in cultured rat hepatocytes. Ethanol-induced decrease of cell viability was inhibited by GABA treatment. Western Blot analysis revealed that GABA inhibits ethanol-induced expression of GRP78 and CHOP, which are UPR markers. These results suggest that GABA is protective against ethanol's cytotoxicity through down-regulation of UPR in cultured hepatocytes.
