三元重金属-农药-离子液体混合物对秀丽线虫的致死协同效应

摘要:离子液体由于具有水溶性和持久性,成为人们关注的污染物。为了探究离子液体和常见污染物重金属、农药以及三元混合物毒性,本文以秀丽线虫为模式生物,以24 h致死率为毒性终点,以重金属(CuSO4)、2种农药(DIC和GLY)、2种离子液体([hmim]Br和[omim]Cl)为混合物组分,构建4个金属-农药-离子液体三元混合物体系,通过均匀射线设计法(UD-Ray)对每个混合物体系设计5条混合物射线,测定各个组分及混合物射线对秀丽线虫的急性致死毒性,以基于置信区间的组合指数来评价各个混合物射线的毒性相互作用。结果表明,重金属-农药-离子液体混合物之间存在明显的协同作用,4个三元混合物体系的20条射线中有11条呈现明显协同效应。其它混合物射线呈现当混合物浓度水平逐渐升高时,毒性相互作用从拮抗向加和直至协同而变化的规律。为了与传统方法比较,也利用基于NOEC的固定浓度比射线法设计了1 条混合物射线,结果表明4个混合物体系中共4条射线的毒性相互作用与均匀设计射线法相似。

The synergistic lethal toxicity of ternary mixture of heavy metal, pesticide and ionic liquid to Caenorhabditis elegans
Abstract:Ionic liquids attract widespread attention as one kind of contamination due to their water solubility and persistance. To investigate the toxicities of ionic liquids, heavy metals, pesicides and their ternary mixtures, four heavy metal - pesticide - ionic liquid mixture systems were structured, thereinto one heavy metal(CuSO4), two pesticides(DIC and GLY), two ionic liquids([hmim]Br and [omim]Cl) were selected as mixture components and each ternary mixture systems were designed into five rays by UD-Ray. The 24-h lethality was selected as the toxicity endpoint and Caenorhabditis elegans were selected as model organism. The acute lethal toxicity of each components and their mixture rays were tested with Caenorhabditis elegans. Toxicological interactions of these mixture rays were studied using the combination-index (CI) integrated with confidence intervals. The results showed the heavy metal - pesticide - ionic liquid mixture systems displayed obvious synergism and there were synergism in 11 rays of 4 ternary mixture systems. The toxicity interaction of other rays changed from antagonism through addition action to synergism with the concentration increasing. To compare with the classic design method, we designed each mixture system into one ray by fixed NOEC concentration ratio ray and the results showed the toxicity interaction of four rays in four mixture systems were similiar with the results of UD-Ray.
