交感神经对种植体周骨结合作用初探

摘要:目的 研究交感神经系统对种植体骨结合的作用。方法 通过化学性失交感神经法建立全身失交感神经动物模型后进行小鼠股骨远端种植植入术,术后2周及4周分别进行HE、Masson和TRAP染色评价骨组织形态学;推出实验评价骨组织生物力学;并进行ELISA检测骨钙素和Ⅰ型胶原交联羧基端肽(CTX-1)的表达。结果 实验组较正常组增加种植体周破骨细胞数目和活性,骨组织结构矿化成熟延迟。同时,推出实验结果显示实验组股骨内种植体最大推出载荷较对照组降低。而ELISA结果表明,实验组CTX-1显著升高,在4周后仍有统计学意义。但其对骨钙素作用没有对CTX-1作用明显。结论 交感神经能够调控破骨细胞形成和吸收,降低种植体周骨结合力,影响种植体周骨结合的稳定性。

Preliminary evaluation of the role of sympathetic system in peri-implant osseointegration
Abstract:This study aims to investigate the role of sympathetic nervous system in implant osseointegration. After chemical sympathectomy established by serial injections of 6-hydroxydopamine, implant surgery was induced on distal femur head. At 2- and 4-week postoperatively, the femur carrying an implant of each mouse was harvested. HE, Masson and TRAP staining were performed to evaluate peri-implant bone tissue histomorphology; Push-out test was carried out to detect bone tissue biomechanics; ELISA test was performed to detect serum osteocalcin and collagen I crosslinked carboxy-terminal peptide (CTX-1). The results showed that, compared with the control group, the experimental group increased the number of osteoclasts and also approved its activity. Delayed mineralization and maturation in trabecular tissue were observed in experimental group. In addition, the maximal push-out force was decreased in the experimental group. The ELISA results showed that CTX-1 was obviously higher in the experimental group after four weeks. However, the level of osteocalcin didn't showed any significant differences between two groups. Chemical sympathectomy could affect peri-implant bone mineralization and maturation, as well as decrease strength of osseointegration, which suggests that sympathetic nerve system play an important role in prei-implant osseointegration.
