伊曲康唑治疗颅内真菌感染胶束给药系统的构建

摘要:目的:本研究旨在为将聚合物胶束递药系统用于抗真菌药物伊曲康唑,设计并优化处方,得到高载药的伊曲康唑聚合物胶束。方法:使用与伊曲康唑相容性较好的两亲性材料PLGA-PEG,通过考察处方组成、投药量、水化介质量对药物包封率以及载药量的影响,进行星点设计-效应面优化。结果:得到最优处方:PEG-PLGA=10 mg,ITZ= 2mg,水化介质量= 2.47ml;包封率为67%,载药量为6.3%。结论:通过处方优化,成功构建高载药量的PEG-PLGA/ITZ聚合物胶束纳米递药系统。

Itraconazole Micelle Drug Delivery System for Treatment of Intracranial Fungal Infection
Abstract:Aim:Our work is to apply micelle drug delivery system to the antifungal drug itraconazole, design and optimize the formula.Method:With the preliminary experiment, PLGA-PEG is found compatible with ITZ; and by single factor tests and central composite design, we optimize the micelle formula.Result: We successfully get the optimal formula: ITZ = 2mg, water = 2.47ml, with a 67% entrapment efficiency and a 6.3% drug loading capbility.Conclusion: By formula optimization, we successfully obtain a micelle drug delivery system of ITZ/PEG-PLGA with high drug loading capibility.
