黄叶银杏愈伤组织诱导及黄酮的产生

摘要:以普通银杏和黄叶银杏叶片为研究材料,诱导愈伤组织并对愈伤组织添加0.1mmol/L和0.5mmol/L硝酸铈,研究不同浓度硝酸铈对两种银杏愈伤组织中黄酮、内酯含量及POD、CAT和PAL活性、MDA含量等的影响。结果表明:0.1mmol/L和0.5mmol/L硝酸铈都促进了黄叶银杏愈伤组织黄酮的产生,且0.1mmol/L处理的愈伤组织中黄酮含量比0.5mmol/L处理的愈伤组织黄酮含量高了近1倍。0.5mmol/L处理的普通银杏愈伤组织中黄酮含量比黄叶银杏愈伤组织中黄酮含量高了1.7倍。普通银杏叶片中黄酮的含量也明显高于嫁接的黄叶银杏叶片中黄酮的含量。黄叶银杏的POD、CAT活性远高于普通银杏,而PAL酶活性和MDA含量却远低于普通银杏。以上结果说明黄叶银杏愈伤组织和普通银杏的生理代谢存在较大的差异。

Callus induction and effect of Ce3+ on content of flavonoids in golden-leaf ginkgo callus
Abstract:The effect of Ce3+ on flavonoids content in callus of ordinary ginkgo and golden-leaf ginkgo were studied. The results showed that flavonoids content in two kind of ginkgo calluses were increased under 0.1mmol/L and 0.5mmol/L nitric acid cerium (Ce(NO3)3). Flavonoids content in golden-leaf ginkgo under 0.1mmol/L Ce(NO3)3 was higher 100% than that of under 0.5mmol/L Ce(NO3)3. But flavonoids content in ordinary ginkgo callus increased 170% than that of golden-leaf ginkgo callus under 0.5mmol/L Ce(NO3)3 treatment. Flavonoid content in ordinary ginkgo leaves was also significantly higher than that of golden-leaf ginkgo leaves. POD and CAT activity in golden-leaf ginkgo callus were significantly higher than that of ordinary ginkgo, but PAL activity and MDA content were lower than that of ordinary ginkgo under 0.5mmol/L Ce(NO3)3 treatment. These above results showed that the significant difference of physiological metabolism was existed in the two ginkgo germplasms.
