CIP2A通过AKT-NF-κB信号通路促进细胞凋亡

摘要:摘要:目的:探讨CIP2A如何调控Bcl-2和Bcl-xL 的表达。方法:选取HBE和A549细胞系进行实验。采用western blot方法检测细胞系中p-AKT的表达量。使用AKT抑制剂后检测细胞中p-AKT、p-IκBα、Bcl-xL和Bcl-2的表达量,并通过荧光素酶报告基因检测NF-κB转录活性。使用NF-κB特异性抑制剂处理后检测p-IκBα、Bcl-2和Bcl-xL的表达量。结果: CIP2A 促进细胞中Bcl-2和Bcl-xL表达上调而且AKT和IκBα磷酸化活性增加。加入AKT或NF-κB信号通路抑制剂处理后,细胞中Bcl-2以及Bcl-xL表达下调。 结论:CIP2A通过AKT-NF-κB信号通路上调细胞凋亡蛋白Bcl-2和Bcl-xL的表达。

CIP2A inhibits cell apoptosis through AKT-NF-κB pathways
Abstract: Objective The study is to find out how CIP2A regulates the expression of Bcl-2 and Bcl-xL. Method HBE and A549 cell lines were used for research and PDTC and LY294002 were employed in the experiment to detect the expression of Bcl-2、Bcl-xL 、p-AKT and p-κBα by western blot assay . Transcription activity of NF-κB was analyzed through luciferase report essays. Results CIP2A upregulates the expression of p-AKT,Bcl-2,Bcl-xL and p-IκBα, which was reversed by AKT/NF-κB inhibitor. Conclusions CIP2A promotes cell apoptosi through AKT- NF-κB pathways.
