丹参酮ⅡA对斑马鱼胚胎急性毒性的影响

摘要:目的 以斑马鱼为模型,研究丹参酮IIA (Tanshinone IIA) 对斑马鱼胚胎发育急性毒性的影响,评价丹参酮IIA对斑马鱼胚胎的毒性。方法 将受精后3 hpf (hours past fertilization) 的斑马鱼胚胎分别暴露于含有不同浓度的丹参酮IIA试液中,在显微镜观察不同浓度丹参酮IIA对斑马鱼胚胎发育情况的影响,并且统计斑马鱼胚胎的孵化率与致死率,评价丹参酮IIA对斑马鱼胚胎发育的毒性效应。结果 与对照组相比,在浓度≤3 μM丹参酮IIA作用96 hpf内对斑马鱼胚胎无明显毒性效应;在浓度≥6 μM ,丹参酮IIA作用时间48 hpf后,斑马鱼胚胎孵化率显著下降,死亡率增加并产生大量畸形胚胎。结论 丹参酮IIA的浓度≥ 6 μM 的时候对斑马鱼早期生命发育阶段有毒性效应。

Toxic effect of Tanshinone IIA on zebrafish embryo
Abstract: Objective To evaluate the toxicity of Tanshinone IIA on zebrafish embryo. Methods The zebrafish embryoes were expose to a series of different concentrations of Tanshinone IIA. The influence of Tanshinone IIA on zebrafish embryo was observed by Microscope. The hatching rate, mortality rate and malformations of zebrafish embryo were also analyzed. Results Compared with control groups, no obvious toxic effects of less than 3 μM Tanshinone IIA on zebrafish in 96 hpf. When the concentration of Tanshinone IIA was equal or greater than to 6 μM at 48 hpf, embryo exposure to Tanshinone IIA elicited decreased hatching rate, mortality rates and malformations. Conclusion Tanshinone IIA has certain toxicity on the embryonic growth of zebrafish at the concentration equal or greater than to 6 μM.
