猴头菇神经营养活性物质研究

摘要:以发掘更有效的具有神经营养活性的小分子物质为目的,利用层析、谱学和细胞生物学技术对猴头菇子实体中的成分进行了分离,结构鉴定和活性检测。结果在乙醇提取液中的乙酸乙酯段,得到了1个新化合物hericenone K(11)和10个已知化合物,其中4-dihydro-5-methoxy-2-methyl-2-(4'-methyl-2'-oxo-3'-pentenyl)-9(7H)-oxo-2H-furo[3,4-h] benzopyran(10)和11通过手性HPLC可以分别拆分为两个对映异构体;化合物ergosterol peroxide(1),cerevisterol(2),3b,5a,9a-trihydroxy-ergosta-7,22-dien-6-one (3), (±)-10, (-)-10和(+)-10显著增强了NGF诱导的PC-12细胞轴突生长活性。该研究表明猴头菇中的多种小分子化合物可以和NGF共同作用表现出神经营养活性,对防治神经退行性疾病的药物发现具有重要理论和实用意义。

Neurotrophic substances from Hericium erinaceum
Abstract:To discover more efficient neurotrophic factors, one new compound hericenone K (11), along with ten known compounds were purified from an ethanol extract of the fruiting bodies of Hericium erinaceum. Combined with chromatographic,spectroscopic and cell biology techniques,the isolation ,characterization and functional assessment have been done. 3,4-dihydro-5-methoxy-2-methyl-2-(4'-methyl-2'-oxo-3'-pentenyl)-9(7H)-oxo-2H-furo[3,4-h] benzopyran (10) and 11 were revealed to be racemic mixtures by CD specturm and separated by chiral RP-HPLC. Ergosterol peroxide (1), cerevisterol (2), 3b,5a,9a-trihydroxy-ergosta-7,22-dien-6-one (3), (±)-10, (-)-10 and (+)-10 in combination with NGF showed a significant increase in neurite-bearing cells compared to NGF-induced PC-12 cells. These maybe considered as a potential source for treatment of neurodegenerative diseases.
