铜催化N-对甲苯磺酰腙与邻炔苯酚的偶联环化反应

摘要:苯并呋喃是很多天然产物及有药物活性的药物和化学产品的核心骨架。本文报道了一种CuBr催化N-对甲苯磺酰腙与邻炔基苯酚偶联环化反应合成苯并呋喃的新方法。该反应经过铜卡宾的转移插入形成了一种二取代的连烯中间体,然后经由铜催化的分子内环化生成苯并呋喃。反应从简单原料出发,对一系列官能团都具有较好的容忍性。

Cu(I)-catalyzed coupling/cyclization of N-tosylhydrazone and o-hydroxy phenylacetylene
Abstract:Benzofurans are important structural motifs of a wide range of natural substances, compounds of pharmaceutical interest and commodity chemicals. In this paper, A general and practical method to synthesize 2-substituted benzofurans and indoles is described. This method employs easily accessible N-tosylhydrazones and o-hydroxy or o-amino phenylacetylenes as substrates. The reaction proceeds through a CuBr-catalyzed coupling-allenylation-cyclization sequence and tolerates a series of functional groups.
