桦木酸抑制地塞米松诱导脾淋巴细胞凋亡的作用

摘要:为了研究桦木酸(Betulinic acid, BA)对脾淋巴细胞凋亡的影响。以地塞米松(Dexamethasone,Dex)建立小鼠体外脾淋巴细胞凋亡模型,利用倒置显微镜、荧光显微镜和电子透射显微镜观察细胞形态和亚细胞结构,琼脂糖凝胶电泳观察DNA的裂解,流式细胞术测定细胞凋亡率等方法检测BA对Dex介导的小鼠脾淋巴细胞凋亡的影响。实验结果表明,2.5 μmol Dex使脾淋巴细胞凋亡明显增加,染色质发生边聚、浓缩,凋亡小体等典型的凋亡特征,BA明显降低Dex引起的脾淋巴细胞凋亡。这表明BA对Dex诱导的淋巴细胞凋亡具有预防性的保护作用。

In vitro inhibitory effect of betulinic acid on dexamethasone induced splenocyte apoptosis
Abstract:In order to investigate the effect of betulinic acid (BA) on splenocyte apoptosis. Splenocyte apoptosis induced by dexamethasone (Dex) was established in vitro. Morphology and ultrastructure of splenocytes were performed with inverted microscope,fluorescence microscope and transmission electronic microscopy, DNA fragmentation was visualized with agarose gel electrophoresis, and the percentage of apoptosis was measured with flow cytometry by adding BA into cultured mouse splenocytes in vitro. The results showed that the percentage of splenocyte apoptosis was increased by Dex (2.5 μmol) with the cytoplasm and nuclear shrunk, condensed chromatin and formation of apoptotic body, however, BA reduced Dex-induced splenocyte apoptosis. It was suggested that BA could effectively prevent Dex-induced apoptosis, and protect splenocytes.
